The cofactor requirements of sarcosine oxidase.

چکیده
It has been established that the enzyme, sarcosine oxidase, found in the mitochondria of rat liver and elsewhere (I), catalyzes the following over-all reaction (2, 3) : sarcosine + $02 = glycine + serine + HCHO. The cofactor requirements of this reaction have not been established, although Mackenzie (1) has mentioned unpublished results which seemed to indicate that a folic acid deficiency in rats did not affect the relative amounts of serine and HCHO formed. Weinhouse and Friedmann (4), on the other hand, reported that the amount of radioformic acid formed from radiomethyl sarcosine and excreted in the urine was increased in folic acid-deficient rats. In the present work, the effects of a folic acid deficiency and of INHl upon the sarcosine oxidase activity of rat liver mitochondria were studied.

Studies on a Sarcosine Oxidase of Bacterial Origin
A "sarcosine oxidase" was prepared from a creatinine-decomposing strain of Pseudomonas aeruginosa. The enzyme is inactivated by drying, lyophilization, and dialysis against distilled water. No dialyzable cofactor was found. Optimal activity of the enzyme is reached at pH 7.8. Enzyme activity is directly proportional to enzyme concentration and also to substrate concentration up to the point of ...
متن کاملMolybdenum Cofactor Biology and Disorders Related to Its Deficiency; A Review Study
Background: Molybden, as a vital and essential micronutrient is directly involved in the metabolism of other elements including carbon, sulfur, and nitrogen. Molybdenum alone is not biologically active unless it binds to specific cofactors. Except for the bacterial nitrogenase, which contains molybdenum-Iron complex, molybdenum cofactor (Moco) is considered as the bioactive component placed in ...
متن کاملCrystal Structure of Heterotetrameric Sarcosine Oxidase
Introduction The concentration of serum creatinine is widely used as an indicator of renal function. Sarcosine oxidase (SO) has been used in the enzymatic determination of creatinine in the clinical laboratory. SO has been isolated from various bacterial strains. It was first purified from Corynebacterium sp. U-96 (SO-U96) and shown to be a heterotetrameric enzyme. SO-U96 is a complex, bifuncti...
متن کاملAlteration of L-proline oxidase activity of sarcosine oxidase and a structural interpretation
Sarcosine oxidase (EC 1.5.3.1; sarcosine:oxygen oxidoreductase [demethylating]) is a flavoprotein that catalyses the oxidative demethylation of sarcosine to yield glycine, formaldehydethe, and hydrogen peroxide. This enzyme is involved in the bacterial metabolism of creatinine with the related enzymes, creatininase and creatinase. It is industrially important and is used with creatininase and c...
متن کاملThe effect of aminoguanidine on the oxidation of formaldehyde by rat liver.
Formaldehyde was shown to be a normal metabolite when Handler, Bernheim, and Klein (1) found sarcosine oxidase which catalyzes the oxidative demethylation of sarcosine to glycine and formaldehyde. Sakami (2), Welsh and Sakami (3), and Stekol et al. (4) have recently presented evidence that formate may provide the animal with necessary methyl groups. It was therefore of interest to investigate t...
